6-OXO is a steroidal aromatase inhibitor known as suicidal inhibitor as it irreversibly binds to the aromatase enzyme (an enzyme that aromatizes androgens and creates estrogen). Once aromatase is inhibited, it follows that estrogen is minimized leading to a boost in test.

4-AT is a potent irreversible aromatase inhibitor that inhibits estrogen biosynthesis by permanently binding and inactivating aromatase in adipose and peripheral tissue. Aromatase is responsible for the conversion of test to estradiol. Blocking aromatase causes the body to decrease in levels of estradiol, which then results in increase of LH and consequently, test. Since test has myotropic activity and estradiol does not, elevated test levels increase muscle mass.
