About ACE-031
Ace-031, also known as ACVR2B, is a soluble form of activin type IIB receptor, which is an inhibitor of myostatin and other naturally occurring proteins that limit muscle growth. This compound was genetically developed to bind to myostatin before it can attach to its receptor and halt muscle growth. The two Biopharma companies that worked on this compound are Acceleron and Shire.
How does Ace 031 Work?
ACE-031 acts by binding to proteins that normally signal through a particular activin receptor called ActRIIB. Myostatin and other related proteins signal through this receptor to limit muscle growth. When ACE-031 binds to these proteins.
ACE-031 prevents them from interacting with the ActRIIB receptor, thus allowing muscle to grow. The critical difference between ACE-031 and antibodies to myostatin is that ACE-031 inhibits multiple regulators of muscle mass in addition to myostatin.
In preclinical studies, the end result is a greater increase in muscle mass than can be achieved by blocking myostatin alone. In Duchenne, the goal is to offset the loss of muscle due to the lack of dystrophin by increasing the production of new muscle.
